چکیده

Cells can contact the extracellular matrix (ECM) through a variety of specialized structures. These cell-matrix contacts contain membrane proteins such as integrins, which bind both matrix components and cellular proteins, thus bridging the substratum and the cytoskeleton (reviewed in Adams, 2001). Matrix contacts provide the physical linkage that enables cells to adhere to and migrate on (or through) the matrix. Moreover, they can also act as localized ‘relay stations’ for important signaling pathways. Well-known cellmatrix contacts include focal complexes and focal adhesions, whereas others, such as fibrillar adhesions, podosomes and invadopodia are just beginning to attract widespread attention.

The formins FHOD1 and INF2 regulate inter- and intra-structural contractility of podosomes.

Podosomes are actin-rich adhesion structures that depend on Arp2/3-complex-based actin nucleation. We now report the identification of the formins FHOD1 and INF2 as novel components and additional actin-based regulators of podosomes in primary human macrophages. FHOD1 surrounds the podosome core and is also present at podosome-connecting cables, whereas INF2 localizes at the podosome cap struct...
